Partec PA PloidyAnalyzers can be used for Ploidy determinations which are formulated by the precise measurement of the total DNA content of individual nuclei, which can be derived from almost any plant tissue. The PloidyAnalyzer measures the DNA content of many thousands of individual nuclei within seconds or minutes by measuring the relative fluorescence of the total DNA. The result is displayed as a DNA histogram in real time on a large LCD screen. PA-I uses built-in personal computer with software for measuring, analyzing results and reports.

Working Principles:
A flow cytometer is an instrument for counting and analysing particles and cells. The partecPloidy Analyser (PA) is a flow cytometer which is specially developed for ploidy determination on plants. The ploidy levels of the cells (and the numerical ratio between them) will be determined by measuring the nuclear DNA content of the plant cells. With the PA once can measure the DNA content automatically, rapidly and with very high accuracy of chromosome difference. By absolute counting the exact concentration i.e the cell number per ml can be determined. The sample flows through a capillary into the flow cuvette. Because of the specific flow cuvette geometry the sheath and sample current are speeded up. The cells or particles labelled with fluroscent coloring pass the measuring area one after the other. The cells or particles are individually illuminated by the excitation light and the fluorescent light is measured and analysed.

User Instructions:
Sample speed must be fitted to sample concentration. For a very low concentration increase speed. For high measuring accuracy a low speed value of about 0.4ul/s is advisable
